Why does the leucine dose matter?
Leucine is the trigger amino acid for muscle protein synthesis — but a trigger is not the whole mechanism. The dose question, answered plainly.
What was studied
Dose-response research on leucine and essential amino acids: how the muscle protein synthesis response changes with the leucine content of an amino acid dose.
What it showed
The synthetic response scales with leucine availability, and roughly 2.5-4 g of leucine inside a complete EAA mixture robustly triggers it in most adults. Leucine alone cannot sustain synthesis — the other essential amino acids are the building material the trigger spends.
What this evidence does not show
Threshold estimates vary with age, body mass, and the protein source studied. A trigger dose in a study is an average response, not a guarantee for any individual.
